BBL™ Ninhydrin Reagent Droppers are used in the determination of the hippurate reaction to aid in the identification of certain bacteria. The hydrolysis of sodium hippurate is a qualitative test useful in the identification of Streptococcus agalactiae (group B β-hemolytic streptococci), Campylobacter jejuni, Gardnerella vaginalis, Listeria sp. and other aerobic bacteria.1-4 The presence of the enzyme hippurate hydrolase is visualized by the purple color formed when ninhydrin reagent oxidizes the amino acid produced during hippurate hydrolysis. The hydrolysis of sodium hippurate is a qualitative test useful in the identification of Streptococcus agalactiae (group B β-hemolytic streptococci), Campylobacter jejuni, Gardnerella vaginalis, Listeria sp. and other aerobic bacteria.1-4 The presence of the enzyme hippurate hydrolase is visualized by the purple color formed when ninhydrin reagent oxidizes the amino acid produced during hippurate hydrolysis. Examine the reagent for signs of deterioration (see “Product Deterioration”). A reagent control should be performed daily or with each batch of tests and when new 1% sodium hippurate solution is made. The reagent control is tested using the procedure above except that no organism is added. There should be no purple coloration after the 10 min incubation period. When tested according to the recommended procedure, Streptococcus agalactiae ATCC™ 27956 should give a positive reaction and Enterococcus faecalis ATCC 19433 should give a negative reaction. Quality control requirements must be performed in accordance with applicable local, state and/or federal regulations or accreditation requirements and your laboratory's standard Quality Control procedures. It is recommended that the user refer to pertinent NCCLS guidance and CLIA regulations for appropriate Quality Control practices. Ninhydrin Reagent Droppers contain 0.5 mL of 3.5% ninhydrin in 50% aqueous ethanol. For in vitro Diagnostic Use. Ninhydrin is an irritant. Avoid contact with the skin and eyes. If contact occurs, immediately wash skin well with soap and water or flush eyes with copious amounts of water. Follow proper laboratory procedures in handling and disposing of infectious materials. | |||||||||||
