Métodos de análisis
Los métodos de análisis de microorganismos específicos pueden comenzar con una fase de reanimación de varias horas, por ejemplo, un análisis de bacterias Gram-negativas tolerantes a ácidos biliares seguido de uno o dos pasos no selectivos o selectivos de enriquecimiento de microorganismos presentes en una muestra. Posteriormente, se realiza un subcultivo en un medio de agar selectivo para la detección de microorganismos específicos. Siempre se debe confirmar la identidad de los microorganismos que han crecido en el medio en la fase final del análisis.
Los análisis son cualitativos, es decir, determinan la presencia o ausencia del microorganismo específico. Sin embargo, además del análisis cualitativo, se ha descrito un método de análisis cuantitativo basado en el principio del número más probable (véase el análisis de grado de contaminación microbiana) para bacterias gram-negativas tolerantes a ácidos biliares
Los medios deben responder a los requisitos descritos en la farmacopea con respecto a las propiedades de fomento del crecimiento, la selectividad y la electividad, mediante inóculos de bajo tamaño (≤ 100 UFC) de un panel de referencia de microorganismos bajo condiciones de incubación específicas.

Método de extensión en placa

Método del número más probable (NMP)
Para este método, se asume la distribución normal de microorganismos en soluciones acuosas. Existen diferentes variaciones de este método. Sin embargo, la base de todas las variaciones es la misma. El número más probable en la muestra se puede calcular en función de la probabilidad de la presencia de microorganismos viables en varias diluciones de una muestra. En resumen, se preparan diez diluciones de una muestra. De cada dilución se inoculan partes de 1-mL por triplicado en un medio de caldo. Tras la incubación, se inspeccionan todos los caldos para observar la presencia de crecimiento microbiano y se registran los resultados. Normalmente se utilizan tablas estadísticas basadas en la ley de Poisson para leer el número más probable. Las versiones alternativas del análisis emplean más tubos inoculados por dilución, lo que aumenta la sensibilidad.

Recogida activa y pasiva de muestras de aire
El método de filtración por membrana se suele emplear para los materiales de muestra solubles y acuosas. Este método resulta especialmente útil para las muestras que presentan propiedades antimicrobianas. Se filtra un volumen medido de muestra a través de una membrana de filtro estéril con un tamaño de poro nominal no superior a 0,45 μm para retener los microorganismos. Si es necesario, la membrana puede enjuagarse una vez o varias veces para eliminar posibles sustancias inhibidoras que presentes en el material de la muestra. Las soluciones de aclarado más utilizadas son 1 g/L de solución neutra de peptona de carne o caseína (pH 7,1 ±0,2), fluido A, D y K. La solución de aclarado puede contener también agentes neutralizantes o de inactivación. Tras el aclarado, la membrana se transfiere a un medio de crecimiento microbiológico sólido, seguido de la posterior incubación. Se cuenta el número de unidades formadoras de colonias presente en la membranas y se calcula asi el de la muestra. Filtración por membrana
El método de filtración por membrana se suele emplear para los materiales de muestra solubles y acuosas. Este método resulta especialmente útil para las muestras que presentan propiedades antimicrobianas. Se filtra un volumen medido de muestra a través de una membrana de filtro estéril con un tamaño de poro nominal no superior a 0,45 μm para retener los microorganismos. Si es necesario, la membrana puede enjuagarse una vez o varias veces para eliminar posibles sustancias inhibidoras que presentes en el material de la muestra. Las soluciones de aclarado más utilizadas son 1 g/L de solución neutra de peptona de carne o caseína (pH 7,1 ±0,2), fluido A, D y K. La solución de aclarado puede contener también agentes neutralizantes o de inactivación. Tras el aclarado, la membrana se transfiere a un medio de crecimiento microbiológico sólido, seguido de la posterior incubación. Se cuenta el número de unidades formadoras de colonias presente en la membranas y se calcula asi el de la muestra. La mayoría de las empresas utilizan equipos de Millipore (Milliflex®) o Sartorius (Combisart®).
