Membrane Filtration
The membrane filtration method is widely used for aqueous and soluble sample materials. It is particularly useful for samples that exhibit antimicrobial properties. A measured volume of sample is filtered through a micro-organism retaining sterile membrane filter with nominal pore size not greater than 0.45 μm. If necessary the membrane may be rinsed once or several times to eliminate inhibitory substances that were present in the sample material. Commonly used rinsing solutions are 1 g/l neutral solution of meat or casein peptone (pH 7.1 ±0.2), fluid A, D and K. The rinsing solution may also contain neutralizers or inactivating agents. After rinsing, the membrane is transferred onto a solid microbiological growth medium, followed by subsequent incubation. The number of colony forming units (CFU) present on the membrane are then counted, and the number of CFU in the sample is then calculated. Most companies use equipment from Millipore (Milliflex®) or Sartorius (Combisart®).

Most Probable Number (MPN) Method
Normal distribution of micro-organisms in aqueous solutions is an assumption made for this method. There are several variations on this method. However, the basis of all variations is identical. The most probable number in the sample can be calculated based on the probability of presence of viable micro-organisms in several dilutions of a sample. Briefly, ten-fold dilutions of a sample are prepared. From each dilution 1-ml portions are inoculated in triplicate into a broth medium. After incubation all broths are inspected for the presence of microbial growth and results are recorded. Commonly, statistical tables based on Poisson’s law are used to read the most probable number. Alternative versions of the test employ more tubes to be inoculated per dilution, which increases the sensitivity.

Spread Plate Method
A measured volume, usually 0.1 ml up to 0.5 ml, of a (diluted) sample is brought onto a solid medium and evenly spread over the agar surface with a sterile Drigalski spatula. After incubation, the number of colonies present on the agar surface are counted, and the number of CFU in the sample is then calculated.

Direct inoculation
A measured volume of sample is brought aseptically into two media containers, e.g. bottles. Usually, it is necessary to test samples from one batch separately since the volume of the product may not be more than 10 per cent of the volume of the medium. Neutralizers or inactivating agents may be added to the media in case inhibitory factors are present in the product. Then, samples are incubated for 14 days. In case media become turbid because of the nature of the product (suspensions), a subculture into fresh medium is prepared after the first incubation, and the samples are incubated for at least another 4 days

Active & Passive Air sampling
The membrane filtration method is widely used for aqueous and soluble sample materials. It is particularly useful for samples that exhibit antimicrobial properties. A measured volume of sample is filtered through a micro-organism retaining sterile membrane filter with nominal pore size not greater than 0.45 μm. If necessary the membrane may be rinsed once or several times to eliminate inhibitory substances that were present in the sample material. Commonly used rinsing solutions are 1 g/l neutral solution of meat or casein peptone (pH 7.1 ±0.2), fluid A, D and K. The rinsing solution may also contain neutralizers or inactivating agents. After rinsing, the membrane is transferred onto a solid microbiological growth medium, followed by subsequent incubation. The number of colony forming units (CFU) present on the membrane are then counted, and the number of CFU in the sample is then calculated. Most companies use equipment from Millipore (Milliflex®) or Sartorius (Combisart®).

Test methods
Methods for the test for specified micro-organisms may start with a resuscitation stage of several hours, e.g. test for bile-tolerant Gram-negative bacteria, followed by one or two non-selective or selective enrichment steps of microorganisms present in a sample. Subsequently, subculturing on selective agar media for the detection of the specified micro-organism is performed. The identity of micro-organisms grown on the media in the final stage of the test should always be confirmed.
Test are qualitative, i.e. presence or absence of the specified micro-organism. However, in addition to the qualitative test, a quantitative test method based on the MPN principle (see bioburden testing) has been described for bile-tolerant Gram-negative bacteria.
Media should meet the requirements described in the pharmacopoeia with respect to growth promoting properties, selectivity and electivity, using low size inocula (≤ 100 CFU) of a panel of reference micro-organisms under specified incubation conditions.

Surface Sampling
Surface sampling methods include contact plating and swabbing. Contact plates are useful for sampling flat, smooth surfaces. In addition, locations should be well accessible. Swab sampling is useful for irregular surfaces, nooks, gaps, crevices and other difficult to access locations. Surface sampling is also used for sampling garments and gloves. Results are expressed as numbers of CFU per plate (contact plates) or cm2(swabs). Media residues should be removed adequately after sampling.

Pour Plate Method
A measured volume, usually 0.5 ml up to 2 ml, of a (diluted) sample is brought into a sterile Petri dish. Subsequently, molten agar medium (< 45°C) is added and mixed with the sample and left until solidified. After incubation, the number of colonies mainly present in the agar and some on the agar are then counted, and the number of colony forming units (CFU) in the sample is then calculated.
